Sirolimus

Sirolimus is an immunosuppressive agent that inhibits T-lymphocyte activation and proliferation. It is primarily used to prevent renal transplant rejection, often in combination with other immunosuppressive drugs, and to treat lymphangioleiomyomatosis, a rare progressive lung disease.

Dosage by Body Weight

Dosage must be highly individualized and managed by a specialist. Standard adult dosing starts with a loading dose of 6 mg followed by 2 mg daily, adjusted based on therapeutic drug monitoring (target blood trough concentrations). Pediatric dosing in children 13 years of age or older (weighing less than 40 kg) is calculated based on body surface area (1 mg/m² daily after a 3 mg/m² loading dose).
