Leflunomide

Leflunomide is an oral disease-modifying antirheumatic drug (DMARD) with immunomodulatory and anti-inflammatory properties. It works by inhibiting dihydroorotate dehydrogenase, an enzyme essential for pyrimidine synthesis, which in turn limits the proliferation of autoimmune T-lymphocytes responsible for joint inflammation.

Dosage by Body Weight

For adults, therapy is sometimes initiated with a loading dose of 100 mg once daily for 3 days, followed by a maintenance dose of 10 mg to 20 mg once daily. Due to risk of side effects like hepatotoxicity, many clinicians omit the loading dose and start directly with the maintenance dose of 10 mg to 20 mg daily.
